Cell Biomaterials, 2026, 2, 100455.<\/em><\/p>\n<p>&nbsp;<\/p>\n<p>Clinical translation of pluripotent stem cell-based therapies has been hampered by difficulties in fabricating and implanting dense, metabolically active tissues at the human scale. By combining sacrificial 3D printing with polymer dip coating to produce vascular lattices housed inside ready-made pouches, this study develops a practical approach to deliver therapeutic cells with immediate convective perfusion rather than relying on slow host vascular ingrowth.\u00a0<em>In vitro<\/em>\u00a0perfusion experiments show rapid, physiologically relevant insulin secretion and stem cell-derived islet maturation. Week-long arteriovenous shunt implants in pigs demonstrate safety and patency under blood flow. As an alternative to diffusion-limited encapsulation devices, this strategy proposes off-the-shelf bioartificial organs that can be manufactured, sterilized, stored, filled with cells at the point of care, and directly anastomosed to the host blood circulation.
